Juventus, the four-time defending champion, was eliminated from the Coppa Italia on Wednesday with a 3-0 quarter-final defeat by Atalanta, while AS Roma suffered a humiliating 7-1 defeat by Fiorentina.
Juventus were looking for a fifth straight title in the League Cup and Cup Cup this season.
But a doubling of Duvan Zapata after the first match of Timothy Castagne allowed the Bergamo team to meet in the semifinals with Fiorentina.
Juventus Cristiano Ronaldo won his first trophy of the season in Italy's Super Cup this month.
But they suffered their first defeat in national competition this season, while they go up high and are unbeaten in Serie A.
Mbadimiliano Allegri's players had come from behind against Lazio in the league this weekend, with Ronaldo taking a 2 to 1 victory in Rome.
But the Portuguese superstar failed to score his first goal in Italy's Cup. Juventus, also touched by defender Giorgio Chiellini, is injured in the first half-hour.
Castagne opened the scoring after 37 minutes and Zapata resumed two minutes later, before adding a third to four minutes from the end.
The Colombian ace also scored a 2-2 double in the last meeting between the two teams in Serie A on 26 December.
"It's only human to not always be on top," said Allegri, who was banned from the sideline because of dissent.
"I do not know how many games the guys have won in Rome we had a terrible first half, but we fought back, and tonight everything went wrong.
"It would have been crazy to think we could win every game, it hurts to go out because we really wanted this trophy.
"Atalanta deserved his victory, they were better and showed their mental strength."
– "Sorry sorry sorry" –
Earlier in Tuscany, Federico Chiesa scored a hat-trick as Fiorentina crushed a "shameful" team from Roma.
Chiesa scored two goals early in the game, adding a third in the second half, overtaking Roman goalkeeper Robin Olsen. Giovanni Simeone scored twice in the last ten minutes.
Colombian Luis Muriel and Marco Benbadi also found the bottom of the net against a team from Rome who felt bad after losing three goals ahead of a 3-3 draw against Atalanta during the Serie A weekend. AT.
"I am bitter, but I will not resign," said Roma coach Eusebio Di Francesco.
"I am disappointed with the team, there are no excuses.
"I have a hard time explaining what happened, because it was a game we were mistaken in any way imaginable. It was shameful."
Di Francesco's team struggled after a difficult start to the season with four straight wins in Serie A and Coppa Italia, and are now fifth in the championship.
"What's frustrating is that it's so inexplicable," said the Roma boss. "Whenever we think we are cured, we discover that the disease is still there."
Chiesa scored the first goal after seven minutes from a Cristiano Biraghi center.
Bryan Cristante's head shook the post before Chiesa, 21, scored his second goal on Olsen shortly after a quarter of an hour.
Aleksandar Kolarov reduced the score in Rome after half an hour, but Fiorentina had two quick goals over Muriel.
The Colombians created Benbadi after 66 minutes, while Simeone left the bench to do more harm to Roma, who ended the match with ten men after second-half substitute Edin Dzeko was sent off for a second yellow card.
"It's the hardest day of my career," said Roma sports director Monchi.
"I have never seen anything like it and we can only apologize to all the fans of Roma, I can only repeat, sorry, sorry, sorry."
New rookie Krzysztof Piatek scored a brace on Tuesday as AC Milan beat Napoli 2-0 at San Siro.
Milan, winner five times, will face either Inter Milan or Lazio, who will play Thursday, on two runs in the last four.
