The Congress of Trade Unions warned Theresa May that she was about to throw her weight behind calls for a referendum on the final Brexit deal.
TUC leader Frances O 'Grady said that unless the government concludes "an agreement the workers need" with the EU, it would require a "popular vote".
"It's only fair that people have a say," she told the BBC.
The government ruled out another EU referendum, saying it was working to succeed Brexit.

Labor says they're not asking for another public vote, but that a referendum on Brexit conditions should be "on the table".
A referendum like this is the demand for the popular vote campaign, which is supported by some MPs from the two largest parties.
Speaking at the BBC's Andrew Marr parade at the start of the TUC conference in Manchester, Ms. O. Grady urged the government to extend the UK's membership in the EU to allow more negotiations.
"Time is running out and a collapse of the EU would be an absolute disaster for the people we represent," she said.
She said the livelihoods of companies like BMW, Airbus and Jaguar were at stake, calling for the creation of jobs and workers' rights as a government priority.
"I want to inform the Prime Minister today that if we do not get the agreement the workers need, then the TUC will put all our weight behind a campaign for a popular vote so that the people have their say the agreement is good enough or not, "she said, adding that union leaders always consult with their members when they negotiate an agreement.
Last week, the GMB union announced its support for the Popular Vote campaign.
To coincide with the start of the TUC conference, People's Vote published polls suggesting that most members of Unison, Unite and GMB supported the claims of the campaign.
The YouGov poll of more than 2,700 trade unionists was evidence of the "growing momentum" of his campaign.
